Researchers have identified previously unknown immune structures within the skull bone marrow that act as a local brain defense center. The study, published in Nature, could lead to new treatments for brain tumors and neurological conditions.

Scientists have discovered a new immune organ hiding inside the skull that may play a significant role in protecting the brain. In a study published in 2026 in the scientific journal Nature, researchers from the Washington University School of Medicine in St. Louis identified previously unknown immune structures in the bone marrow of mouse skulls. Experiments demonstrated that these structures help the immune system fight cancerous brain tumors and that enhancing their activity can improve therapeutic outcomes.
Despite centuries of medical research, scientists continue to uncover structures in the body that were previously unknown. This discovery is located in a particularly surprising place—inside the skull, in close proximity to the brain. The research team identified an immune organ that acts as a local defense center.
Brain Defense Mechanism
The researchers found areas where immune system cells gather, multiply, and prepare to deal with threats. Their location allows them to respond to problems occurring in the brain without relying solely on immune cells arriving from other parts of the body.
Jonathan Kipnis, a researcher of the immune system and the brain at Washington University and senior author of the study, explained: "This research reveals that the skull bone marrow is much more than just a structural frame—it contains previously unknown centers for immune responses unique to the brain." According to him, "uncovering this local immune environment changes the way we view the connections between the nervous system and the immune system, and opens up new and exciting directions for treating brain tumors and other neurological diseases."
This new organ functions like a security station for the immune system. In the body, there are areas where immune cells undergo a process that prepares them to fight specific threats. While such structures are typically known from lymph nodes, researchers have now identified similar structures within healthy skull bone marrow.
Impact on Tumor Treatment
To understand the significance of the discovery, the team examined mice with glioma. By applying a gel under the scalp, they either weakened or strengthened the activity of these immune centers. The results showed a clear correlation: when the activity of these structures was disrupted, tumors became more aggressive. Conversely, when the researchers used a combination of three proteins to stimulate immune activity, the anti-cancer response was strengthened, significantly increasing the survival rate of the mice.
It is important to note that this discovery was made in mice. However, the presence of follicular T-cells in human skull bone marrow suggests that a similar defense system may exist in humans. Further research is required to confirm this and understand its functionality.
If the organ is found to operate similarly in humans, it could revolutionize treatment. Instead of invasive procedures directly on brain tissue, future therapies might target these immune centers through the skull or scalp to boost the immune system's attack against tumors. This could potentially lead to new treatments for conditions including Alzheimer's, Parkinson's, schizophrenia, and long COVID.
